Development of antioxidant rich dietary supplement for celiac disease

ABSTRACT

Celiac Disease (CD) is an immune reaction caused due to intake of gluten protein called gliadin in susceptible individuals. Studies had shown the correlation between celiac disease and an increase in ROS (Reactive Oxygen Species) in the body which resulted in oxidative stress in people with celiac. Glutathione, a natural antioxidant, present in the body could be regenerated with diet rich in antioxidants. This research aims to alleviate the oxidative stress caused by the ingestion of gluten in celiac patients. It focuses on developing plant-based antioxidant-rich powdered dietary supplements from medicinal plants such as Emblica Officinalis, Zingiber Officinalis, Curcuma longa, Terminalia chebula, Piper longum, Azadirachta indica and Foeniculum vulgare, that has the ability to reduce the oxidative stress and makeup the nutrients deficiencies in Celiac patients. The Free Radical Scavenging Assay using DPPH (1, 1-Diphenyl-2-picrylhydrazyl) method and protein estimation using Lowry’s method were used to analyse the antioxidant activity and the protein concentration of the powdered sample respectively.IC50 value was calculated to be 3.81 mg/ml, which showed strong antioxidant activity of the powdered sample. The protein content of the concoction was found to be 1.625 mg/ml for 1 ml of the sample concentration. This study is limited to analysis of antioxidant activity using DPPH assay and estimation of protein using Lowry’s method due to paucity of resources. The testing of digestion of gluten protein by the powdered sample and the effectiveness of the powdered product in celiac patients is excluded for the time being.
